Do bleed nipples have to point up?

I'm installing the big brake kit from Ireland, and one of the caliper's nipples point up, while the other points down. Looks like two of the same side. Do the nipples really have to point up?

If they don't point up you have to remove the calipers from the strut and bleed them while holding them upright (bleed nipples pointing up).

You might try flicking it gently with your fingertips, or teasing it with an ice cube...
